When people think of the most bike-friendly city in the US, most people are thinking of Minneapolis, Minnesota.
Some may think of Portland, Oregon, though.

The League of American Bicyclists list over 100 communities in there Bicycle Friendly Communities Program. They are ranked into four categories. This is probably the most systematic assessment of bicycle friendliness for cities in the USA. The highest ranking communities are Boulder, Davis and Portland. The is more information in the links.
Bicyling Magazine recently released a list with the 50 best cities to bike in. The first city was Minneapolis, Minnesota. They based the list on many factors including: segregated bike lanes, municipal bike racks, bike boulevards, cyclist involvement in local government, a vibrant and diverse bike culture, and smart savvy bike shops. They also accounted for geographic diversity so California would not dominate the list.
