The EPA is the largest, government run environmental organization with nearly 20,000 full time employees. Other, citizen groups like the Sierra Club are much larger, with member bases in the millions, but they do not have the same authority in environmental regulation that the EPA does. The Sierra CLub and other groups can only lobby and try to influence the decisions of the EPA.
Actually, I think the United Nations Environment Programme probably has the most far-reaching positive effect on the environment, even withstanding the weakness of the resolutions its member countries have thus-far passed.
