Can bookstores load onto e-reader for someone with no computer skills (and no computer!)?

That depends on the bookstore. I don’t think that a small traditional bookstore would have the knowledge base to do it, but you might be able to find someone who can help you at a larger bookstore that actually sells e-readers. Since Barnes and Noble sells e-readers, you might try your luck with them. The best way to know for sure it to walk into the store and ask there!

If the employees are unable to help you, chances are you will run into someone there reading an ereader (especially if there is a coffee shop in the bookstore).  You can always ask them to either show you how or at least give you a few tips!
